The successful candidate will participate in studies to determine the molecular mechanisms regulating cancer development and to develop targeted therapeutics that prevent immune suppression during cancer. These studies evaluate two key areas of cancer: 1) How a receptor for common environmental pollutants (i.e. the aromatic hydrocarbon receptor/AHR) regulates cancer metastasis and survival, and 2) how the AHR suppresses tumor-specific immune response. Some of the work will involve AHR inhibitors which we postulate will both inhibit tumor survival and enhance tumor immunity. Approximately half of the work will involve DNA manipulation, subcloning, transfection, gene editing (CRISPR/Cas), gene reporter assays, western blotting, and maintaining tumor cell lines in culture. Much of the remaining half will involve mouse studies in which mouse tumors are transferred to naïve recipients or mouse studies in which primary tumors are generated in vivo with carcinogen. In both cases, we will be evaluating the ability of recently developed drugs (AHR inhibitors) to enhance tumor immunity. The successful candidate will also perform a modicum of basic laboratory tasks including preparing and aliquoting reagents, maintaining cell culture stocks, maintaining the nitrogen cell storage tank, ordering supplies, and logging experimental procedures. Required Skills The successful candidate should have significant previous laboratory experience and be familiar with if not skilled in: animal (mouse) work including intraperitoneal injections, flow cytometry, RNA and DNA extraction, qPCR, gene cloning, gene reporter assays, western blotting, mammalian cell culture, freezing and thawing cell lines, and preparing culture media, buffers, and stock solutions. The successful candidate is expected to be a competent and careful investigator who is capable of independent work and who is interested in joining an enthusiastic and supportive team of investigators dedicated to developing novel strategies for human cancer treatment. Co-authorship on publications is likely for the competent and dedicated research assistant. High School Diploma/GED Required. Bachelor’s Degree Preferred. Less than 1 Year of experience required.
